明膠是(shi)一(yi)種非常重要(yao)的(de)天(tian)然生(sheng)物高分子材料,是(shi)由動物的(de)皮骨及(ji)結(jie)締組(zu)織中的(de)膠原部分降解(jie)成的(de)。在(zai)食(shi)品、化(hua)妝品和制(zhi)藥工業被廣泛使用。明膠一(yi)加熱即(ji)成為液體,遇冷則變成彈性固體凝膠塊。膠體的(de)溶液-凝膠互相轉(zhuan)換是(shi)明膠特有的(de)現(xian)象(xiang)。因(yin)此,形成凝膠的(de)能力(li)是(shi)明膠一(yi)項很重要(yao)的(de)性能。很早就有人(ren)試圖通過測算,用數字表示(shi)凝膠強度(du)。
最早(zao)的(de)評估(gu)凝(ning)膠強度(du)的(de)方法是(shi)完全依靠人的(de)“指(zhi)測(ce)”。“指(zhi)測(ce)”方法是(shi)用手指(zhi)簡單按(an)壓一下明(ming)膠表面,再與已知標準明(ming)膠的(de)抗力進(jin)行(xing)對比。這種方法雖然(ran)迅速,但是(shi)數據可靠性(xing)遠遠不(bu)及科學測(ce)量。再加上人體感官器(qi)官的(de)靈(ling)敏(min)性(xing)不(bu)夠。很難通過指(zhi)測(ce)進(jin)行(xing)精(jing)確(que)、客觀(guan)的(de)評估(gu)。
國標GB6783-2013 <<食品添加劑(ji) 明膠>> 中(zhong)有(you)關(guan)于凝膠強(qiang)度的測(ce)定(ding)(ding)方(fang)法。配(pei)置指(zhi)定(ding)(ding)濃度的明膠溶(rong)液(ye),經過(guo)靜置、水浴加熱攪(jiao)拌溶(rong)解、冷水浴等樣品溶(rong)液(ye)制(zhi)備(bei)過(guo)程后,取放于特(te)定(ding)(ding)的樣品杯(bei)內(nei)。選用特(te)定(ding)(ding)的凝膠測(ce)試探頭,對樣品杯(bei)中(zhong)心點進行穿刺測(ce)試。從(cong)而測(ce)得凝膠強(qiang)度數值。
歐洲藥典EP8.0中(zhong)也采(cai)用相(xiang)同的(de)方法(fa)進行測(ce)定。具(ju)體方法(fa)如下:
- 柱形探頭:12.7±0.1 mm
- 樣品:明膠溶液濃度為6.67%,10℃下
- 樣品瓶規格:59±1 mm(內徑)* 85 mm(高度)
- 測試中速度:0.5 mm/s
- 刺入深度:4 mm
- 測試結果:達4 mm時力值即為凝膠強度。
- 測試方法:
取(qu)(qu)7.5 g待測(ce)樣品(pin)(pin)放入(ru)樣品(pin)(pin)瓶(ping)內,加(jia)入(ru)105 ml的水(shui),加(jia)蓋靜止1-4 h后,在65±2℃水(shui)浴加(jia)熱15 min ,水(shui)浴加(jia)熱過程(cheng)中(zhong)攪(jiao)拌。室溫(wen)下冷卻(que)15 min,加(jia)膠(jiao)塞將(jiang)樣品(pin)(pin)瓶(ping)于10.0 ±0.1℃下冰浴17±1 h。擦干樣品(pin)(pin)瓶(ping)外壁(bi)的水(shui)后進行凝膠(jiao)強度測(ce)定,測(ce)定點(dian)為(wei)樣品(pin)(pin)表面的中(zhong)心點(dian)。取(qu)(qu)兩(liang)組的平(ping)均值作為(wei)報告值。
